IGF-1 LR3 is a modified analog of Insulin-Like Growth Factor-1 (IGF-1) with an arginine substitution at position 3 and a 13-amino acid extension that reduces its affinity for binding proteins. This modification extends its half-life from ~15 minutes (natural IGF-1) to ~20–30 hours and increases potency by ~3×.
Mechanism: IGF-1 is the primary mediator of growth hormone's anabolic effects. It binds to IGF-1 receptors on muscle cells, activating mTOR and PI3K pathways that stimulate protein synthesis, satellite cell proliferation, and muscle hypertrophy. LR3 modification reduces binding to IGF-binding proteins (IGFBPs), keeping more IGF-1 free and active.
